Opis:
Mice infected with 200 Trichinella spiralis larvae were killed at 3-69 days post infection (dpi) and the jejunum and masseter muscles were sectioned in a cryostat and examined in the Tuneli method with "In situ Cell Detected Kid POD" of Boehringer-Mannheim. Data concering the exact localization and dynamie of apoptic cells in both organs are presented. The authors conclude that apoptosis plays a important role in the pathogenesis of trichinellosis.

Administration intraperitoneally of the Ascaris suum extract - ASE-(0.6-1.4 g of Ascaris proteins/kg/day) at a late stage of organogenesis (8-12 days of gestation) disturbed course of mouse pregnancy. lt has been found that injections of higher doses of ASE to pregnant mice caused the symptoms manifesting maternal toxicity (decreased body weight gain /p < 0.001/ as compared to control, intrauterine resorption of litter, vaginal hemorrhages, female mortality and altered behaviour). There is a linear interrelationship between the logarithm of the dose of ASE and mortality of pregnant mice. The DL₅₀ value of Ascaris proteins for pregnant mice was 1.02 g/kg/day (confidence interval 0.97-1.07 g/kg/day). ASE exerted embryotoxic effects: significantly decreased the number of surviving fetuses per litter and the mean body weight of fetuses, increased the number of fetal resorptions.

Administration intraperitoneally of the Ascaris α-chymotrypsin inhibitor (40-300 mg/kg/day.) at a late stage of organogenesis (8-12 days of gestation) disturbed course of mouse pregnancy. The low doses of α-chymotrypsin inhibitor (40-80 mg/kg/day) significantly decreased the number of live fetuses per litter, increased the number of fetal resorptions. The symptoms of matemal toxicity that occurred after administration of the highest doses of the inhibitor (80-300 mg/kg/day) to pregnant mice included: decreased body weight gain as compared to control, vaginal hemorrhage, intrauterine resorption of litters, abortions, altered behaviour of animals immediately after injection and death. There is a linear interrelationship between the logarithm of the doses of the inhibitor and mortality of pregnant mice. The DL50 value of the inhibitor for female was 116 mg/kg /day (confidence interval: 95.5-140.0 mg/kg/day).

Opis:
Behaviour of macrophages in experimental mice trichinellosis, was investigated using the immunoenzymatic technique with monooclonal antibodies CD11b/CD18 within the framework of avidin-biotin-DAB. The maximum and earliest mobilization of macrophages 7 day post infection (dpi) was observed in the lamina propria of the intestinal mucosa. The highest level of macrophages in the muscles tissue was noted on day 21 of infection, however as early as in 28 dpi, their maximum level was observed inside the larval capsules. They line, especially between 35-42 dpi internal capsule surphace.

The influence of the some immunomodulators (PHA-P, TFX and dexamethasone) on the process of apoptosis, occurring in the course of trichinellosis in mice, has been studied. It has been found that PHA-P activates this process in the jejunum mucosa and prolongs it in the muscular inflammatory infiltration, whereas TFX has no influence and dexamethasone distinctly decreases the level of the apoptotic cells. The number of the intestinal trichinae on the successive days of infection was similar in all groups of animals, however, the number of the muscular larvae in the groups receiving immunostimulators was much lower and in the group treated with dexamethasone - a little higher than that in control. As in mice receiving PHA-P and TFX, the cellular inflammatory infiltration in the muscles was larger than that in control, and in the group to which dexamethasone was administrated - smaller, the authors think that it was extensiveness of the infiltration and not the level of the apoptotic cells that influenced the number of the outliving larvae.

In the course of trichinellosis in mice the numerous cells inside larval capsules were observed. Beginning on 14 day post infection (dpi) they were seen in about 7% of infected muscles fibers but at 21 dpi the percentage of capsules with the cells amounted to 77. In the next stage of infection the number of capsules containing cells continued to increase and reach almost 100% at 60 dpi. The identification of the cells was carried out by the indirect immunoenzymatic method using anti-CD4⁺, anti-CD8⁺ and anti-macrophage (Mac-1 antigen) monoclonal antibodies. In course of observation the level of CD4⁺ cells decreased from 11.3 to 0.7, of CD8⁺ from 17.7 to 3.0 and of macrophages from 71.0 to 23.1 %. The most numerous CD4⁺ cells were seen at 14 dpi while the CD8⁺ and macrophages at 21 dpi. Beginning on 28 dpi negatively reacting cells inside larval capsules were also observed. The number of these cells increased from 23.8 to 73.2% at 60 dpi and their identification requires further investigations. The role and the function of all these cells are discussed.

Opis:
Rodents, as mice and rats are the most common laboratory animals used in research and testing. They are seldom investigated for autochthonous ecto- and endoparasites prior their utilization in the experiments. Helminth parasites can alter the interpretation of final results. Pinworms commonly infecting laboratory rodents include mainly the mice pinworms Syphacia obvelata and Aspiculuris tetraptera, and in rats Syphacia muris. The fact that many laboratory rodent colonies were found to be parasite contaminated suggests a need for eradication and improvment of the quality of laboratory rodents. This review reports the data on the presence of helminth parasites in laboratory rodents colonies, and suggests to pay special attention on controlling the sanitary conditions of animal houses.

Studies were undertaken to evaluate by parasitological techniques the course of intestinal and muscular invasions in mice infected with Trichinella spiralis and treated with TFX-Thymomodulin (TFX-Th). Studies were conducted on 120 mice of BALB/c and SWISS strains, infected each with a mean invasive dose of 300 T. spiralis larvae. TFX-Th (Thymoorgan GmbH Pharmazie Co. KG, Vienenburg, Germany) was administered subcutaneously at 30 mg/kg body weight. Depending upon duration of TFX-Th administration and stages of intestinal and muscular invasions, three experiments were distinguished in the study. The results indicated that TFX-Th promoted eradication of larvae from muscles, whether administered at the early of the late stage of T. spiralis invasion.
