Opis:
This paper presents a qualitative and quantitative chromatographic analysis (GC/MS) of the lipophilic fraction extracted from leaves, inflorescences and rhizomes of Silphium perfoliatum L. collected in 1998-2000. It was shown that the extracts of leaves and inflorescences are similar in composition, containing among their main constituents: α-amyrine (up to 13.9% of total components in leaves, up to 11.3% in inflorescences), heptacosane (up to 7.1% in leaves, up to 7.6% in inflorescences), stigmasterol (up to 8.1% in leaves, up to 2.2% in inflorescences), γ-sitosterol (up to 6.9% in leaves, up to 2.2% in inflorescences), β-amyrine (up to 4.1% in leaves, up to 5.8% in inflorescences), β-caryophyllene (up to 2.6% in leaves, up to 1.5% in inflorescences), caryophyllene oxide (up to 4.4% in leaves, up to 2.9% in inflorescences), germacrene D (up to 13.8% in leaves, up to 9.7% in inflorescences) and α-pinene (up to 3.3% in leaves, up to 2.4% in inflorescences). The chemical composition of lipophilic extract of rhizomes significantly differs from that of leaves and inflorescences. The following compounds can be counted among the dominant ones occurring in rhizome extracts: diterpene of labdane type - 16-acetoxycarterochaetol (up to 45%) and sesquiterpenes - 7-b-H-silphiperfol-5-ene (up to 9.1%), 7-a-H-silphiperfol-5-ene (up to 10.7%), d-elemene (up to 7.6%), isocomene (up to 5.6%), germacrene D (up to 5.2%) and modhephene (up to 5.1%).

Opis:
This study presents a protocol for a fast and effective in vitro axenic culture of Huperzia selago (Huperziaceae Rothm.) sporophytes, a club moss which is a source of huperzine A, an alkaloid of a considerable therapeutic potential extensively investigated for its uses as treatment for some neurodegenerative diseases. The proposed procedure allowed approximately tenfold shortening of the species developmental stages with the omission of the gametophyte stage while the sporophyte mass could be increased tenfold within a 6-month period. The cultures were established using vegetative propagules (bulbils) procured from sporophytes growing in the wild without degrading the habitats of this endangered plant species. Explants underwent surface and internal disinfection to eliminate the epiphytic and endophytic bacteria and fungi. In in vitro cultures, the optimum results were achieved using Moore (Mr) medium without growth regulators or supplemented with 0.015 mg/l IBA and 0.3 mg/l kinetin. These media ensured both viability of the propagules and their further development. The biomass growth index for H. selago sporophytes grown from propagules, determined at 3 months of culture (1 passage) on Mr medium with IBA and kinetin was 650%. At 6 months, the biomass growth index increased to 1114%. Vigorous growth of adventitious roots, especially on Mr medium with the addition of 0.25 mg/l NAA, and callus formation on shoot apices were observed. At 6 months of culture, some sporophytes obtained from the bulbils were used as the initiating material for shoot subcultures, which developed best on Mr medium with IBA and kinetin.

Multiple shoots of Telekia speciosa were cultivated on MS medium containing 4.44 µM BAP and 0.54 µM NAA, solidified with agar. After eight weeks of culture the shoots were harvested and extracted with methanol. From the methanol extract one pseudoguaianolide – 2,3-dihydroaromaticin and three thymol derivatives: 8-hydroxy-9,10-diisobutyryloxythymol, 10-isobutyryloxy-8,9-epoxythymyl isobutyrate and 10-(2-methylbutyryloxy)-8,9-epoxythymyl isobutyrate were isolated as major secondary metabolites. Moreover, the shoots produced megastigmane and monoterpene glucosides, which were isolated for the first time from the species. The content of 2,3-dihydroaromaticin in the shoot culture was similar to that found in the intact plant, whereas yields of the three thymol derivatives were higher from multiple shoots than from the plants grown in the open field.

Visual chromatography has been employed for a preliminary identification of natural compound of a group of sesquiterpene lactones – Centaurea bella Trautv., C. crocodylium L., C. lusitanica Boiss. et Reuter, Helenium hoopesii A. Gray = Dugaldia hoopesii (A. Gray) Rydb., Stizolophus balsamita (Lam.) Cass., Zoegea baldschuanica C. Winkl., germacranes – Santolina pinnata Viv. subsp. neapolitana (Jord. et Fourr.) Guinea = S. neapolitana Jord. et Fourr. A dependence of the colour of the spots, induced by anisaldehyde reagent, on the presence of several substituents in the germacranolide ring – derivatives of partnenolide and salonitenolide – has been identified. The structures of the skeleton in the ring of sesquiterpene lactones in Helenium hoopesii can also be established by thin layer chromatography (TLC). The analysis of the chromatograms of extracts from dry and fresh Centaurea crocodylium herb has shown significant differences as for the chemical composition. Two sorts of germacranes of Santolina pinnata subsp. neapolitana display a characteristic colour of the spots.
