S100A4 is a member of the S100 family of calcium-binding proteins that is directly involved in tumor metastasis. In the present study, we examined the potential role of S100A4 in metastasis in breast cancer and its relation with matrix metalloproteinase-13 (MMP-13). Analysis of 100 breast cancer specimens including 50 with and 50 without lymph node metastasis showed a significant upregulation of S100A4 and MMP-13 expression in metastatic breast cancer tissues. Positive immunoreactivity for S100A4 was associated with MMP-13 expression. Overexpression of S100A4 in the MDA-MB-231 breast cancer cell line upregulated MMP13 expression leading to increased cell migration and angiogenesis. SiRNA-mediated silencing of S100A4 downregulated MMP13 expression and suppressed cell migration and angiogenesis. Moreover, neutralization of MMP-13 activity with a specific antibody blocked cell migration and angiogenesis in MDA-MB-231/S100A4 cells. In vivo siRNA silencing of S100A4 significantly inhibited lung metastasis in transgenic mice. The present results suggest that the S100A4 gene may control the invasive potential of human breast cancer cells by modulating MMP-13 levels, thus regulating metastasis and angiogenesis in breast tumors. S100A4 could therefore be of value as a biomarker of breast cancer progression and a novel therapeutic target for human breast cancer treatment.

Opis:
Calorie restriction (CR) is known to extend life span from yeast to mammals. To gain an insight into the effects of CR on growth and development of the silkworm Bombyx mori at protein level, we employed comparative proteomic approach to investigate proteomic differences of hemolymph and fat body of the silkworm larvae subjected to CR. Thirty-nine differentially expressed proteins were identified by MALDI TOF/TOF MS. Among them, 19 were from the hemolymph and 20 from the fat body. The hemolymph of the CR group contained two down-regulated and 17 up-regulated proteins, whereas the fat body contained 15 down-regulated and five up-regulated ones. These proteins belonged to those functioning in immune system, in signal transduction and apoptosis, in regulation of growth and development, and in energy metabolism. Our results suggest that CR can alter the expression of proteins related to the above four aspects, implying that these proteins may regulate life span of the silkworm through CR.

Opis:
Poly(L-lactic acid) (PLLA) as an important biodegradable polymer suffers from slow crystallization rate and poor heat resistance. An organic compound N,N’-dodecanedioic bis(3-phenylpropionic acid) dihydrazide (BHADD) was synthesized to evaluate its general influences on the physical properties of PLLA. The melt-crystallization process indicated that BHADD could serve as a heterogeneous nucleating agent for improving the crystallization of PLLA, and PLLA/1%BHADD exhibited the sharpest melt-crystallization peak located at the highest temperature, as well as an increase of cooling rate weakened the crystallization ability of BHADD-nucleated PLLA. And the final melting temperature also displayed the significant effect on the crystallization process of PLLA. For the cold-crystallization process, both BHADD concentration and heating rate affected the cold-crystallization behavior of PLLA/BHADD, the increasing of BHADD concentration caused the cold-crystallization peak to shift to the lower temperature; in contrast, a higher heating rate during heating leaded to the peak’s shift toward the higher temperature because of the thermal inertia. The melting behavior of PLLA/BHADD depended on the crystallization temperatures and heating rates, and the double melting peaks were attributed to the melting-recrystallization. Thermal decomposition experiment showed all PLLA/BHADD samples as the pure PLLA only exhibited one thermal decomposition stage, but PLLA/BHADD had a lower thermal stability than the pure PLLA. Additionally, the addition of BHADD seriously decreased the light transmittance of PLLA.
Poli(kwas L-mlekowy) (PLLA) to ważny biodegradowalny polimer charakteryzujący się małą szybkością krystalizacji i słabą odpornością na ciepło. Dihydrazyd kwasu bis(3-fenylopropiono) N,N'-dodekanodiowego (BHADD) zsyntetyzowano w celu oceny jego wpływu na właściwości fizyczne PLLA. Przebieg procesu krystalizacji ze stopu świadczy o tym, że BHADD może służyć jako heterogeniczny środek zarodkujący zwiększający szybkość krystalizacji PLLA. Mieszanina PLLA/1% mas. BHADD wykazywała najostrzejszy pik krystalizacji ze stopu zlokalizowany w zakresie najwyższej temperatury, a jednocześnie większą szybkość chłodzenia osłabiającą zdolność krystalizacji PLLA zarodkowanej BHADD. Również końcowa temperatura topnienia w istotnym stopniu wpływała na proces krystalizacji PLLA. W procesie krystalizacji na zimno zarówno stężenie BHADD, jak i szybkość ogrzewania oddziaływały na zachowanie PLLA/BHADD podczas krystalizacji, zwiększenie stężenia BHADD powodowało przesunięcie piku krystalizacji na zimno w kierunku niższej temperatury, natomiast większa szybkość ogrzewania prowadziła do, spowodowanego bezwładnością cieplną, przesunięcia piku krystalizacji na zimno w kierunku wyższej temperatury. Zachowanie PLLA/BHADD podczas topnienia zależało od temperatury krystalizacji i szybkości ogrzewania, a drugi pik topnienia przypisano procesowi rekrystalizacji. Badany przebieg rozkładu termicznego świadczy, że wszystkie próbki PLLA/BHADD, tak jak czysty PLLA, wykazywały tylko jeden etap rozkładu termicznego, mieszanina PLLA/BHADD charakteryzowała się jednak mniejszą stabilnością termiczną niż czysty PLLA. Dodatek BHADD w znacznym stopniu zmniejszył przepuszczalność światła PLLA.

Serpins are a broadly distributed family of protease inhibitors. In this study, the gene encoding Bombyx mori serpin-2 (Bmserpin-2) was cloned and expressed in E. coli. The Bmserpin-2 cDNA contains a 1125 bp open reading frame (ORF). The deduced protein has 374 amino-acid residues, contains a conserved SERPIN domain and shares extensive homology with other invertebrate serpins. RT-PCR analysis showed that Bmserpin-2 was expressed in all developmental stages of B. mori larvae and various larval tissues. Subcellular localization analysis indicated that Bmserpin-2 protein was located in the cytoplasm. Interestingly, real-time quantitative PCR revealed that the expression of Bmserpin-2 in the midgut of susceptible B. mori strain 306 significantly increased at 72 hours post inoculation (hpi) when infected with BmNPV. However, there was no significant increase of the Bmserpin-2 expression in resistant strain NB infected with BmNPV. Thus, our data indicates that Bmserpin-2 may be involved in B. mori antiviral response.

Opis:
Human cytomegalovirus (HCMV) UL4 and UL5 genes are two members of the RL11 gene family. In an earlier study, three UL4 transcripts of about 1.7, 1.5 and 1.4 kb were found in early and late classes after infection by the Towne strain by nuclease protection and primer extension analyses. In the present study, two UL4 transcripts (1.5 and 1.7 kb) were found by cDNA library screening, Northern blot, 3' and 5' RACE analyses to appear initially in the immediate early phase and one UL4 transcript (1.4 kb) in the late phase in a low-passage clinical isolate. Furthermore, two novel low-abundance UL5 transcripts with the same 3' terminus as the identified UL4 transcripts in the UL4-UL5 gene region were found in late class RNAs.

For a 2-connected cubic graph $G$, the perfect matching polytope $P(G)$ of $G$ contains a special point \( x^c = ( \tfrac{1}{3},\tfrac{1}{3},…,\tfrac{1}{3}) \). The core index $ \phi(P(G)) $ of the polytope $P(G)$ is the minimum number of vertices of $P(G)$ whose convex hull contains $ x^c$. The Fulkerson’s conjecture asserts that every 2-connected cubic graph $G$ has six perfect matchings such that each edge appears in exactly two of them, namely, there are six vertices of $P(G)$ such that $ x^c $ is the convex combination of them, which implies that $ \phi(P(G)) \le 6 $. It turns out that the latter assertion in turn implies the Fan-Raspaud conjecture: In every 2-connected cubic graph $G$, there are three perfect matchings $M_1$, $M_2$, and $M_3$ such that $M_1 \cap M_2 \cap M_3 = \emptyset $. In this paper we prove the Fan-Raspaud conjecture for $ \phi(P(G)) \le 12 $ with certain dimensional conditions.

Opis:
To achieve enhanced accuracy of fabric representation and defect detection, an innovative approach using a sparse dictionary with small patches was used for fabric texture characterisation. The effectiveness of the algorithm proposed was tested through comprehensive characterisation by studying eight weave patterns: plain, twill, weft satin, warp satin, basket, honeycomb, compound twill, and diamond twill and detecting fabric defects. Firstly, the main parameters such as dictionary size, patch size, and cardinality T were optimised, and then 40 defect-free fabric samples were characterised by the algorithm proposed. Subsequently, the Impact of the weave pattern was investigated based on the representation result and texture structure. Finally, defective fabrics were detected. The algorithm proposed is an alternative simple and scalable method to characterise fabric texture and detect textile defects in a single step without extracting features or prior information.

Opis:
An up-flow constructed wetland (UFCW) incorporating a novel membrane-less air-cathode single-chamber microbial fuel cell (MFC) was designed to treat dye wastewater and simultaneously generate bioelectricity. The performance of UFCW-MFC was evaluated via Methyl Orange (MO) and chemical oxygen demand (COD) removal rates and the output voltage. For comparison, the performance of a single UFCW was also assessed. A repeatable and stable voltage output of about 0.44±0.2 V was obtained in UFCW-MFC. The MO and COD removal rates in UFCW-MFC were 93.5 and 57.2%, respectively, significantly higher than those in single UFCW (75.4 and 42.6%, respectively), suggesting the obvious enhancement of electrodes on MO and COD removal. The anode zone of UFCW-MFC made the most contribution to MO and COD removal compared with other layers. The oxidation-reduction potential (ORP) and dissolved oxygen (DO) profiles showed that the anaerobic environment was well developed in the lower part of UFCW-MFC (0–24 cm) and the upper part (41–42 cm) had a good aerobic environment, thus greatly contributing to the MO anaerobic reduction and aerobic degradation of breakdown products. These results obtained here suggest that the UFCW-MFC may provide an effective alternative for the treatment of dye wastewater and simultaneous bioelectricity generation.

Cre recombinase from bacteriophage P1 is widely used in both in vitro and in vivo DNA manipulations. Based on a structural and functional analysis, three deleted cre mutants were constructed and expressed in Escherichia coli. Mutated recombinases were purified and their recombination activities were determined in vitro. Our results revealed that the mutant with amino-terminal deletion retains the recombination activity as high as wild type Cre; however, the carboxy-terminal deletion and the middle region deletion both lead to a complete loss of the recombinase function.

Opis:
Azo dye wastewater treatment is urgent necessary nowadays. Electrochemical technologies commonly enable more efficient degradation of recalcitrant organic contaminants than biological methods, but those rely greatly on the energy consumption. A novel process of biofilm coupled with electrolysis, i.e., bioelectrochemical system (BES), for methyl orange (MO) dye wastewater treatment was proposed and optimization of main influence factors was performed in this study. The results showed that BES had a positive effect on enhancement of color removal of MO wastewater and 81.9% of color removal efficiency was achieved at the optimum process parameters: applied voltage of 2.0 V, initial MO concentration of 20 mg/L, glucose loads of 0.5 g/L and pH of 8.0 when the hydraulic retention time (HRT) was maintained at 3 d, displaying an excellent color removal performance. Importantly, a wide range of effective pH, ranging from 6 to 9, was found, thus greatly favoring the practical application of BES described here. The absence of a peak at 463 nm showed that the azo bond of MO was almost completely cleaved after degradation in BES. From these results, the proposed method of biodegradation combined with electrochemical technique can be an effective technology for dye wastewater treatment and may hopefully be also applied for treatment of other recalcitrant compounds in water and wastewater

An attapulgite suspended (AS) filter material was successfully prepared and used in combination with biological aerated filter (BAF) to pretreat excess organic matter and ammonia nitrogen in micropolluted water. The AS filter material was of low density and floated on the water surface, which is beneficial to optimize the hydraulic conditions. Through scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR) It was found that the surface of the filter material was uneven, the pores were deep. When the hydraulic load is 4 m/h and the gas to water volume ratio is 2:1, the removal rates of permanganate index (CODMn) and ammonia nitrogen by ASBAF are up to 57.49 and 88.11%, respectively, and the effluent quality meets relevant standards. After backwashing, the pollutants removal rate will return to stable filtration after two hours. A short-term shutdown of the reactor has little effect on the processing performance of ASBAF, but a long-term shutdown will greatly affect the removal rate of CODMn. The complete organic matter degradation model can well fit the experimental effluent water quality.

W artykule omówiono nowatorskie zastosowanie dynamicznej sieci bayesowskiej (DBN) do oceny niezawodności elektrycznego systemu trakcyjnego ze szczególnym uwzględnieniem metod modelowania DBN. W związku z rosnącą złożonością elektrycznych systemów trakcyjnych oraz wynikającą z niej coraz większą ilością współzależności między komponentami, systemy te narażone są coraz częściej na awarie części składowych. Chociaż istnieje wiele badań dotyczących oceny niezawodności systemów trakcyjnych, stosowane obecnie narzędzia nie mają odpowiedniej mocy modelowania koniecznej do opisu zależności funkcjonalnych i czasowych pomiędzy częściami składowymi. W niniejszej pracy zaproponowano nową metodę modelowania generowania DBN, którą można stosować w odniesieniu do systemów składających się z pewnych określonych komponentów oraz różnych typów rozchodzących się przez nie przepływów. Zależności funkcjonalne i czasowe opisano, odpowiednio, za pomocą tablicy komponentowych prawdopodobieństw warunkowych (Component-based Conditional Probability Table, CPT) oraz tablicy czasowo-zależnych prawdopodobieństw warunkowych. Ponieważ złożoność systemu nie pozwala na zamodelowanie go w prosty sposób jako DBN, do automatycznej budowy modelu DBN wykorzystano algorytm przeszukiwania wszerz (Breadth-First-Search). Oceny niezawodności systemu trakcyjnego z wykorzystaniem proponowanej metody opartej na DBN można dokonywać w dowolnym czasie, co ma ogromne znaczenie przy planowaniu konserwacji w celu zapewnienia bezpieczeństwa systemu.
The article introduces a novel application of a Dynamic Bayesian Network (DBN) in the reliability assessment with regard to the traction system of Electric Multiple Units (EMU), which focus on modeling approach to DBN construction. As a result of high complexity and growing interdependencies, it is increasingly vulnerable to the failure of components. Although many studies on the use of BN for estimating the system reliability have been conducted, there is a lack of effective modeling power regarding current tools in depicting both functional and temporal dependencies between components. In this paper, a new modeling approach to DBN generation is submitted, which can be applied to the system made up of certain components and different types of flows propagating through them. The Component-based CPT (Conditional Probability Table) and Time-dependent CPT are used to describe functional dependencies and temporal dependencies respectively. As the complexity of the system cannot be modeled in a tractable way as a DBN, a Breadth-First-Search (BFS) algorithm is introduced for the construction of the DBN model in an automated manner. With the application of the proposed DBN-based approach, the reliability of the traction system can be evaluated at any given time, which is of great significance to determine the plan of maintenance in an effort to ensure the system safety.

A total-k-coloring of a graph G is a coloring of V ∪ E using k colors such that no two adjacent or incident elements receive the same color. The total chromatic number χ′′(G) of G is the smallest integer k such that G has a total-k-coloring. Let G be a graph embedded in a surface of Euler characteristic ε ≥ 0. If G contains no 3-cycles adjacent to 4-cycles, that is, no 3-cycle has a common edge with a 4-cycle, then χ′′(G) ≤ max{8, Δ+1}.

Opis:
A lightning protection system (LPS) of an urban 110 kV substation is designed and analysed according to NFPA 780 and IEC 62305-3 standards. The analysis of the LPS is established on the value of risk assessment. The total area of the plant is described by one soil layer with uniform resistivity. This study aims to improve the understanding of an unexpected manner of the grounding system beneath lightning currents by clarifying the basic concepts of the lightning protection level and the new design procedure in this paper was clarified according to NFPA-780 level 1 for a lightning protection system. The program is integrated with the CDEGS software, which provides effective geometrical modeling with object and result visualization. Furthermore, module and automated fast Fourier transform (FFT) is implemented in this study to simulate electromagnetic fields in the time and frequency domains. These current values are compared to the desired protection levels within the standards. The study results show that for the improved protection of the system against lightning, the total power grid must be considered as a source of improvement for studying shielding influence and the protection levels provided inside this substation.

Opis:
The aim of the present study was to assess the physicochemical compatibility of a promising energetic salt, 5-amino-1H-tetrazole nitrate (5-ATN), with some typical materials. Thermal techniques (differential scanning calorimetry (DSC) and vacuum stability test (VST)) and non-thermal techniques (X-ray diffractometry (XRD) and Fourier Transform Infrared Spectroscopy (FTIR)) were applied. Five energetic materials (TNT, RDX, HMX, CL-20 and AP) and three common additives (Al, DOS and F2604-2) were tested to evaluate their compatibility with 5-ATN. Based on the DSC results, except for AP that was only partially compatible with 5-ATN, all of the selected materials exhibited good compatible with 5-ATN. The VST test further confirmed the compatibility of the 5-ATN/AP mixtures. Combined with the thermal methods, the FTIR results agreed with the DSC findings. The XRD results showed some differences.
