Modern forest selection programs concern natural and protected populations of trees characterized by a great genetic variability and also production stands frequently deformed by human activity. In the latter, artificial regeneration is prevailing. Forest nurseries working in the organizational structures of State Forests can guarantee the proper provenance (genotype)of seeds and the quality of seedlings obtained from them. The condition, however, is a close uniformity in the conservation of forest gene resources, and selection breeding of forest trees. The success of the program depends also on the development of modern infrastructure for forest nurseries and applied technologies. The latter task was the field of activity of Professor Bolesław Suszka, Ph.D., and his many-year research and organizational and didactic activity in Poland and abroad. The methods in long-term storage and their pretreatment in case of dormant seeds, developed by Professor Suszka, created the theoretical and practical basis for the conservation of forest gene resources not only in Poland.

Opis:
Hladnikia pastinacifolia Rchb., a narrow endemic, has an extremely restricted distribution in Trnovski gozd (Slovenia), despite the presence of many sites with suitable habitats. We compared the morphological traits of plants from different populations and habitats. The overall pattern showed that the smallest plants, with low fruit number, are found on Èaven (locus classicus or type locality); the largest individuals, with high fruit number, grow in the Golobnica gorge. As judged by plant size and seed set, the optimal habitats are screes. We used RAPD markers to estimate genetic variation between and within populations, as well as between and within the northern and the southern parts of the distribution area. Hladnikia showed only a low level of RAPD variability. AMOVA partitioned the majority of genetic diversity within selected populations. The low genetic differentiation between populations and their genetic depauperation indicates survival in situ, since the Trnovski gozd plateau most likely was a nunatak region in the southern Prealps during Pleistocene glaciations. Later range expansion of extant populations was limited by poor seed dispersal. We also analyzed the cpDNA trnL-F intergenic spacer to check whether the sequence is useful for studying the phylogenetic relationships of Hladnikia within the family Apiaceae (Umbelliferae). Our results support the assertion that H. pastinacifolia is an old taxon.

Garlic virus A (GarV-A), Garlic virus B (GarV-B), Garlic virus C (GarV-C) and Garlic virus X (GarV-X) are members of the genus Allexivirus in the family Alphaflexiviridae. In this study, we collected 10, 30, 10 and 14 isolates of GarV-A, GarV-B, GarV-C and GarV-X, respectively, from different parts of Poland. All sequences of coat protein (CP) and nucleic-acid binding protein (NABP) regions of Allexivirus isolates available in GenBank were also included in this study. The nucleotide and amino acid sequences identities within each population differed substantially depending on the region of the genome and virus species. The results of selection pressure analysis showed that populations of each Allexivirus underwent negative selection, but the extent of the negative selection varied. It was also concluded that the GarV-A and GarV-C populations underwent a decrease in population size or balancing selection, while the GarV-B and GarV-X populations underwent an increase in population size. It was concluded that both populations of GarV-X evolved independently in each respective area, in contrast to populations of GarV-A, GarV-B and GarV-C.

Opis:
Application of molecular methods in biology has revolutionized ecology, population genetics and evolutionary biology. For example, the molecular studies revealed complex mating systems and social structures. Analysis of molecular markers revealed the existence of cryptic boundaries such as secondary contact (hybrid zones) among previously isolated populations. Most recently, due to simultaneous study of numerous loci (the population genomics approach), the distribution of estimates of genetic differentiation (Fst) from individual loci has allowed the identification of candidate loci to be under selection. Such analyses may help answer many important evolutionary questions and will also improve inferences in classical population genetics dealing with demography and evolutionary history. Several papers concerning interesting aspects of population genetics and molecular ecology revealed by Polish scientists are discussed and future perspective for population genomics approach are highlighted.

This paper presents results of the population studies of the VNTR polymorphic systems of human DNA. DNA samples were taken from at least 106 unrelated persons living in the Gdansk area. DNA samples were subjected to the PCR amplification and PCR products separated on polyacrylamide gels. Allelic frequencies ofthree AMPFLPs systems and six STRs were calculated and homogeneity of alleles distribution between Gdansk and other population samples were compared. The presented data are the core of the planned population-based DNA indentification system for Poland.

The HLA system is highly polymorphic and commonly used as a marker to understand core population genetical aspects such as stratification, human migration, predisposition to diseases and tissue transplantation compatibility. Although HLA alleles are generally treated as equidistant molecular units in population genetic studies, the diversity of DNA allelic frequencies among populations is also considered to be crucial to interpret the observed HLA polymorphisms. This study compares the variation of HLA DRB1 alleles across various subgroups of both Indian and African Populations, using their respective allelic frequencies. The HLA DRB1 allelic frequencies for 3,153 individuals of 32 subgroups of both Indian and African populations, were collected from the Allele Frequency Net Database (AFND). Correlation matrix and Grid diagram were compiled using the R statistical package. Allele frequencies were analyzed for the respective population subgroups using XLSTAT. The North East Indian populations were completely devoid of HLA DRB1* 16, which is fairly present in south Indian populations. The South Indian populations harbor both HLA DRB1* 10 and HLA DRB1* 15 at relatively higher levels but are very scarce of HLA DRB1* 07, HLA DRB1* 09, HLA DRB1* 12 and HLA DRB1* 13. The alleles HLA DRB1* 03, HLA DRB1* 04 and HLA DRB1* 07 could represent an epicenter for the admixture of African, North-East Indian and South Indian populations.

Multiplex PCR is a useful technique for estimating genetic diversity. This paper presents 3 new sets of primer pairs for effectively amplifying 10 microsatellite DNA loci from rainbow trout (Oncorhynchus mykiss). Unlike other sets of primer pairs that have been developed for amplifying rainbow trout microsatellite loci, ours do not require the hot-start PCR technique. In the paper, we describe the steps taken to choose the loci for each multiplex assay and to verify the genotyping results. We provide the compositions of the PCR mixture and the characteristics of the PCR thermal profile recommended for amplification.

Revolution is just going on. Only correct population genetics stops to be the “only correct”. The information just takes the place of genes. Less serious psychological aspects play here role of main factors. Haldane and Jablonka are symbols of consecutive revolution in the biology.

A sample of Betula nana from the Linie reserve near Dąbrowa Chełmińska, have been fingerprinted using random amplified polymorphic DNA (RAPD). The high level of genetic variation was detected. All individuals had unique genotypes, supporting the generally high resolving power of RAPD’s. For the conservation strategy, information about the distribution of the genetic variation within and among populations plays very important role. Thus, extensive study in other populations of dwarf birch is needed.

The genetic diversity of two Aconitum species endemic to the Carpathian Mountains and Sudetes was studied. A reticulate evolution between them was earlier postulated as an effect of secondary contact. The genetic diversity at the individual and taxonomic levels was examined across the entire geographical ranges of the taxa in 11 populations based on 247 AFLP markers found in 112 individuals in the Sudetes and Western Carpathians. The overall genetic differentiation was greater within the Sudetic A. plicatum (Fst = 0.139, P < 0.001) than within the Carpathian A. firmum (Fst = 0.062, P < 0.001), presumably due to the long-lasting geographic isolation between the Giant Mts and Praděd (Sudetes) populations of the species. Interestingly, relatively distant and presently isolated populations of A. plicatum and A. f. subsp. maninense share a part of their genomes. It could be an effect of their common evolutionary history, including past and present reticulations. The introgression among infraspecific taxa of Aconitum is common, probably as a result of seed dispersal within a distance of ca. 20 km (Mantel's r = 0.36, P = 0.01). Aconitum f. subsp. maninense had the highest genetic diversity indices: Nei's h and rarefied FAr, and divergence index DW (P < 0.05), pointing to its presumably ancient age and long-term isolation.
