Introduction. Treatment of epilepsy patients with one antiepileptic drug often fails and then the insufficiently medicated patients need two or three antiepileptic drugs combined together to stop their seizures. However, polytherapy is always associated with drug-drug interactions whose nature may or may not be favorable for epilepsy patients. Preclinical studies on animals can help to select beneficial combinations of antiepileptic drugs that could be used in further clinical settings. Aim. To isobolographically characterize anticonvulsant effects of a combination of three antiepileptic drugs (carbamazepine, lacosamide and lamotrigine) at the fixed drug dose ratio of 1:1:1 in the mouse maximal electroshock-induced seizure test. Material and methods. Maximal electroconvulsions were evoked in male Swiss mice by a current (25 mA, 500 V, 0.2 s stimulus duration) delivered via auricular electrodes. Type I isobolographic analysis was applied to assess the interaction among carbamazepine, lacosamide and lamotrigine. Results. Isobolographic analysis revealed that the combination of carbamazepine, lacosamide and lamotrigine produced additive interaction in the mouse maximal electroshock-induced seizure test. Conclusions. Additivity among carbamazepine, lacosamide and lamotrigine in this preclinical study can be translated to clinical settings and this three-drug combination can be recommended as a treatment option for epilepsy patients who are resistant to standard treatment regimens.

Opis:
Celem pracy jest opracowanie modeli numerycznych budynków oraz przeprowadzenie analizy wytężenia budynków pod wpływem działania obciążeń wyjątkowych. Na podstawie naziemnego skaningu laserowego, dokumentacji archiwalnej oraz przeprowadzonej inwentaryzacji budowlanej opracowano dyskretne modele obliczeniowe konstrukcji budynków, wykorzystując możliwości programu metody elementów skończonych. Przeprowadzono analizę stanu wytężenia konstrukcji budynków pod działaniem obciążeń wybuchowych. Praca zawiera również analizę zachowania konstrukcji po usunięciu elementów uszkodzonych w wyniku oddziaływania wybuchu ładunku materiału wybuchowego.
The elaboration of the numerical models of building objects is the aim of the paper as well as execution of the analysis of effort of the buildings under the influence of the accidental loadings. Using the possibilities of the program of the finite element method, the discreet computational model of the buildings construction were worked out on the basis of ground-based laser scanning, archival documentation and conducted inventory control. The analysis of the effort state of the construction of buildings for explosive loadings was conducted. The paper also contains the analysis of the construction behaviour after removal of elements damaged in the result of the explosion action of the explosive material charge.

Foresters, considering the character of their work, are exposed to the occurrence of many types of diseases. The main occupational risk among forestry workers is caused by infectious pathogens, the main reservoir of which are wild animals, and the vector are ticks. Forestry workers are exposed to the effect of repellents, including DEET (N,N-diethyl-m-toluamide) in association with protection against tick and mosquito bites. The goal of the project was to discover the amount of DEET identified in the samples of sweat and urine from the foresters who use chemical agents repelling insects. The study covered 22 foresters from the Janow Lubelski Forest District, and 10 individuals who constituted a control group. Questionnaires concerning the use of repellents, and urine samples were collected (2 from each person in the study), as well as sweat samples (4 each). Each collected biological sample was properly prepared and subjected to chromatographic analysis (GC/MS) for the identification of DEET. The season of repellents use is from March–October, or even November. The highest intensity of the use of repellents (as many as 5 days a week) is during the period from May–July. The chromatographic analyses performed did not show even the lowest content of DEET in the collected biological samples from both the control group and foresters. The absence of DEET in the foresters’ urine may be explained by the late date of collection of the samples – in October, when they no longer use repellents, or use them very rarely. The studies conducted within this project allowed the observation that DEET is not accumulated, is subject to quick elimination from the body, which favourably affects the safety of its use. Exposure to chemical occupational hazards in forest areas is an underestimated problem of occupational medicine and public health, which has not been fully explored.
Leśnicy z uwagi na charakter wykonywanej pracy narażeni są na występowanie różnego typu chorób. Głównym zagrożeniem zawodowym dla pracowników leśnictwa są patogeny zakaźne, których głównym rezerwuarem są dzikie zwierzęta, a wektorem kleszcze. Pracownicy leśnictwa są narażeni na działanie repelentów w tym DEET (N,N-dietylo-m-toluamid) w związku z ochroną przed ukłuciami kleszczy i komarów. Celem projektu było poznanie, jaka ilość DEET zostanie zidentyfikowana w próbkach potu i moczu pochodz ących od leśników, którzy stosują środki chemiczne odstraszające owady. W badaniach wzięło udział 22 leśników Nadleśnictwa Janów Lubelski oraz 10 osób stanowiących grupę kontrolną. Zostały zgromadzone ankiety dotyczące stosowania repelentów oraz zostały zebrane próbki moczu (po 2 od każdej osoby badanej) i próbki potu (po 4). Każda z zebranych próbek biologicznych została odpowiednio przygotowana i poddana analizie chromatograficznej (GC/MS) celem identyfikacji w nich DEET. Sezon stosowania repelentów przypada na okres od marca do października, a nawet listopada. Największe natężenie stosowania repelentów (bo aż 5 dni w tygodniu) przypada na okres od maja do lipca. Przeprowadzone badania chromatograficzne nie wykaza ły nawet najmniejszej zawartości DEET w zgromadzonych próbkach biologicznych pochodzących zarówno od grupy kontrolnej, jak i od leśników. Brak obecności DEET w moczu leśników należy tłumaczyć późnym terminem zbiorem próbek – w październiku, kiedy leśnicy nie stosują już repelentów lub stosują bardzo rzadko. Badania przeprowadzone w tym projekcie pozwoliły zaobserwować, że DEET nie jest akumulowany, podlega szybkiej eliminacji z organizmu, co wpływa korzystnie na bezpieczeństwo jego stosowania. Ekspozycja na chemiczne zagrożenia zawodowe na obszarach leśnych jest niedocenionym i nie do końca zbadanym problemem medycyny pracy i zdrowia publicznego.

Introduction and objective. The aim of this study is to assess the interpersonal relations of employees of State Forests in Poland, as well as to indicate the factors related to the level of satisfaction with the relations among people employed in similar positions, and relations with superiors. Materials and method. The issues investigated were developed based on research conducted between 2014–2015 among employees of State Forests in Poland. In each of the 9 forest districts selected, 25 people were selected for testing from 3 groups of employees: foresters, deputy foresters and supervisors, together with administrative staff. Altogether, 399 people took part in the study conducted by means of a questionnaire. Results. The study showed that the overall level of satisfaction with interpersonal relations among employees of State Forests in Poland is high. Analysis indicated that the demographic characteristics of respondents, such as age, gender or place of residence, had no significant influence on satisfaction with interpersonal relations in the workplace. However, the level of satisfaction with interpersonal relations was positively affected by a high level of overall job satisfaction and satisfaction with remuneration. Decrease in the level of satisfaction was influenced by the accumulation of stressful life events and employment in administration. Conclusions. Satisfaction with interpersonal relationships in the workplace is an important indicator of employee mental health. Studies have shown the need to broaden the scope of research into the variables that determine the character and personality of an employee.

Introduction: Anticonvulsant properties of newly synthesized compounds and potential antiepileptic drugs are usually assessed in screen tests in experimental animals. One of the most commonly used screen tests in mice is the maximal electroshock-induced seizure test that reflects tonic-clonic seizures in humans. Materials and Method: A series of 3-p-isopropoxyphenylpyrrolidine-2,5-dione derivatives, including N-aryl and N-arylaminomethyl analogs, were characterized for their anticonvulsant properties in the maximal electroshock-induced seizure test in mice. Electroconvulsions (tonic-clonic seizures) were evoked in adult Albino Swiss mice by a current (sine-wave, 25mA, 50Hz, 500V, 0.2s stimulus duration) delivered via auricular electrodes. Results: N-aryl derivatives did not show any anticonvulsant activity, whereas some representatives of N-arylaminomethyl derivatives, i.e. N-Mannich bases, exhibited a distinct protective action against maximal electroshock-induced (MES) convulsions in mice. Conclusions: Several N-arylaminomethyl derivatives of 3-p-isopropoxyphenylpyrrolidine-2,5-dione may become in future new antiepileptic drugs, or they could serve as valuable supporting materials for obtaining new derivatives with stronger anticonvulsant activities than their maternal compounds.

Introduction and objective: The aim of the study was to determine the effects of 3-(N-p-isopropoxyphenylsuccinimidomethylamino)-cinnamic acid (IPPSMA-CA – a new succinimide derivative) on the protective action of 4 classical antiepileptic drugs (AEDs): carbamazepine [CBZ], phenobarbital [PB], phenytoin [PHT] and valproate [VPA]), against maximal electroshock (MES)-induced tonic seizures in mice. Materials and methods: Tonic hind limb extension (seizure activity) was evoked in adult male albino Swiss mice by a current (sine-wave, 25 mA, 500 V, 50 Hz, 0.2 s stimulus duration) delivered via auricular electrodes. Acute adverse-effectprofiles of the combination of IPPSMA-CA and 4 classical AEDs (CBZ, PB, PHT and VPA) with respect to motor performance, long-term memory and skeletal muscular strength were measured in the chimney, passive avoidance and grip-strength tests, respectively. Results: IPPSMA-CA administered at 150 mg/kg (i.p.) significantly elevated the threshold for electroconvulsions in mice (p<0.01). IPPSMA-CA at doses of 50 and 100 mg/kg, however, had no significant impact on the threshold for electroconvulsions in mice. Nor did IPPSMA-CA (100 mg/kg) significantly affect the anticonvulsant activity of CBZ, PB, PHT and VPA in the MES test in mice. None of the examined combinations of IPPSMA-CA (100 mg/kg, i.p.) with CBZ, PB, PHT and VPA (at their ED50 values from the MES-induced seizure test) affected motor coordination in the chimney test, long-term memory in the passive avoidance task, and muscular strength in the grip-strength test in mice. This indicates no possible acute adverse effects in animals. Conclusions: IPPSMA-CA elevated the threshold for electroconvulsions in mice in a dose-dependent manner. However, IPPSMA-CA at a sub-protective dose of 100 mg/kg did not affect the anticonvulsant action of various classical AEDs in the mouse MES model. Thus, the combinations of IPPSMA-CA with CBZ, PB, PHT and VPA are neutral from a preclinical viewpoint.

Introduction and objectives. The aim of the study was to asses the impact of long-term therapy with the second generation antiepileptic drug levetiracetam (LEV) with arachidonyl-2’-chloroethylamide (ACEA), a highly selective cannabinoid CB1 receptor agoniston the process of neurogenesis in a mouse pilocarpine model of epilepsy (PILO). Additionally, a relationship was established between the treatment with ACEA in combination with LEV, and hippocampal neurogenesis in mouse PILO brain. Materials and method. All experiments were performed on adolescent male CB57/BL mice injected i.p. with LEV (10 mg/kg), ACEA (10 mg/kg) and PMSF (30 mg/kg) (phenylmethylsulfonyl fluoride — a substance protecting ACEA against degradation by the fatty-acid amidohydrolase), pilocarpine (PILO, a single dose 290 mg/kg) and methylscopolamine (30 min before PILO to stop the peripheral cholinergic effects of the pilocarpine, 1 mg/kg). The process of neurogenesis was evaluated after10 days treatment with LEV and ACEA. Results. Obtained results indicated that the combinations of ACEA+PMSF+LEV and ACEA +PMSF increased the total number of total newborn cells compared to the control. Moreover, ACEA+PMSF administered alone and in combination with LEV had a significant impact on neurogenesis increasing the total number of newborn neurons compared to the control group. Neither LEV nor PMSF had a significant impact on the number of proliferating cells and newborn neurons when compared to the control PILO group. In turn, LEV administered alone decreased significantly the number of astrocytes. However, ACEA+PMSF has demonstrated significant increase of astrocytes compare to control mice. Conclusions. These data provide substantial evidence that the combination of LEV+ACEA significantly increases the level of newborn neurons in the PILO dentate subgranular zone.

Introduction and objective. One of the main factors determining the burden of breast cancer is the effectiveness of measures taken to combat this cancer including urban-rural differences. The aim of the study was to analyse the differences in breast cancer incidence and disease stage distribution among urban and rural women in the Świętokrzyskie Province as a part of a strategy for breast cancer control. Materials and method. The study of disease stage distribution included 483 female residents of Świętokrzyskie Province who were diagnosed in 2013 with invasive breast cancer, and reported to Świętokrzyskie Office for Cancer Registration. Urban-rural differences in breast cancer incidence in 2002–2013 were presented using Range Ratio (RR). Changes in incidence trends in urban and rural areas were analysed using joinpoint models. Annual Percentage Change (APC) of the rates was calculated for each time trend. Results. Breast cancer incidence rate in the urban female population was higher than in rural women with RR, amounting to 1.43. However, the analysis of trends showed that the pace and direction of change were developing negatively among inhabitants of rural areas. In 2002–2013, in rural women, the age-standardized rate (ASR) values increased by 2.8% per year (p<0.05). The course of ASR trends showed statistically significant urban-rural differences (p=0.004). Analysis of urban-rural differences in disease stage distribution revealed a non-significantly higher proportion of cases with localised stage in urban than in rural areas, amounting to 51.0% and 43.9%, respectively. Conclusions. Existing health inequalities indicate the need to intensify activities in rural areas and should be the starting point for making key decisions in combating breast cancer.
