We present and review the results of fluorescence upconversion and photon echo experiments, and $ab initio$ calculations performed in our group within the last few years with respect to the light harvesting process in purple bacteria. Carotenoids transfer energy to bacteriochlorophyll (BChl) mainly via the carotenoid S$\text{}_{2}$ → BChl Q$\text{}_{x}$ pathway on a ~100 fs timescale. This transfer is reasonably reproduced by considering the Coulombic coupling calculated using the transition density cube method which is valid at all molecular separations. Carotenoids may also serve a role in mediating B800 → B850 energy transfer in LH2 by perturbing the transition density of the B850 as shown by ab initio calculations on a supermolecule of two B850 BChls, one carotenoid and one B800 BChl. Further calculations on dimers of B850 BChl estimate the intra- and interpolypeptide coupling to be 315 and 245 cm$\text{}^{-1}$, respectively. These interactions are dominated by Coulombic coupling, while the orbital overlap dependent coupling is ~20% of the total. Photon echo peak shift experiments (3PEPS) on LH1 and the B820 subunit are quantitatively simulated with identical parameters aside from an energy transfer time of 90 fs in LH1 and ∞ in B820, suggesting that excitation is delocalized over roughly two pigments in LH1. 3PEPS data taken at room and low temperature (34 K) on the B800-B820 suggest that static disorder is the dominant mechanism localizing excitation in LH1 and LH2. We suggest that the competition between the delocalizing effects of strong electronic coupling and the localizing effects of disorder and nuclear motion results in excitation in the B850 and B875 rings being localized on 2-4 pigments within approximately 60 fs.

We demonstrate a way to conjugate a light-harvesting complex, peridinin-chlorophyll-protein, with silver nanowires using biotin-streptavidin linker. In the case of conjugated structure we observe slight increase of the fluorescence intensity of the chlorophyll emission followed by the gradual decrease of the intensity due to photobleaching. For a non-conjugated mixture of peridinin-chlorophyll-protein with silver nanowires only the photobleaching takes place. The results suggest a possible way to fabricate hybrid nanostructures comprising light-harvesting complexes and metallic nanoparticles for achieving the efficient plasmon-induced enhancement of absorption of the light-harvesting complexes.

Organic solar cells have drawn intense attentions in recent years due to their inherent advantages. But the relatively low power conversion efficiency is the main obstacle in the way of organic solar cell commercialization. One of the main reasons that limit the power conversion efficiency is the mismatch between electrical transmission properties and light absorption properties in an organic active layer. In this work, a highly efficient light trapping scheme with a hybrid microlens array is proposed to resolve this contradiction. This structure can achieve broadband absorption enhancement in the spectrum of interest by chromatic aberration correction and hole parameter adjustment. And the light trapping element can be separated from cells to avoid direct contact with an organic layer that may cause electrical defects. Moreover, it is also compatible with low cost manufacturing technologies.

In this work we demonstrate confocal fluorescence imaging of hybrid nanostructures composed of silver nanowires and peridinin-chlorophyll-protein light-harvesting complexes. The length of silver nanowires, which reaches 10 μm, allows for determination of the nanowire position and consequently direct correlation with the fluorescence image. In this way we probe the influence of plasmon induced electromagnetic field on the fluorescence of light-harvesting complexes. When the nanowires are spaced from the light-harvesting complexes by a 10 nm thick dielectric layer, we observe a fluorescence enhancement, which depends upon the laser excitation wavelength. The measured enhancement values are 2.5 and 1.9 for 405 nm and 485 nm, respectively. Larger enhancement for the 405 nm excitation is attributed to direct creation of plasmon excitations in the silver nanowires.

We show that single peridinin-chlorophyll a-protein light-harvesting complexes from dinoflagellate Amphidinium carterae placed near to silver nanoparticles show strongly enhanced fluorescence emission. Single molecule spectroscopy experiments performed at room temperature point toward an enhancement of more than an order of magnitude for optimal conditions. Irrespective of the enhancement, we observe no effect of the metal nanoparticle on the fluorescence emission energy of the complex. This result provides a way to control the optical properties of biomolecules via plasmon excitations in metal nanoparticles.

Chiral properties of peridinin-chlorophyll-protein (PCP) light-harvesting complexes are studied in terms of vibronic dimer theory previously applied to study certain structural aspects of α-crustacyanin pigments. On the base of CD spectra it is shown that the peridinin dimer acts as a chiral group in PCP complexes and its geometrical structure is such that the peridinin monomers cannot be coplanar. Certain observations concerning the energy transfer process in PCP complexes in vivo are also made.

Vibrational dynamics of the excited state in the light-harvesting complex (LH1) have been investigated by femtosecond stimulated Raman spectroscopy (FSRS). The native and reconstituted LH1 complexes have same dynamics. The ν1 (C=C stretching) vibrational mode of spirilloxanthin in LH1 shows ultrafast high-frequency shift in the S1 excited state with a time constant of 0.3 ps. It is assigned to the vibrational relaxation of the S1 state following the internal conversion from the photoexcited S2 state.

Hybrid nanostructures are often composed of inorganic parts and "biological" ones. Optimized through million years of evolution light harvesting proteins are hard to mimic synthetically. Promising strategy in search for efficient solar cells is an attachment of selected natural protein systems to inorganic quantum dots. Such experimental hybrid structures should have improved charge separation properties. Among the most promising proteins is peridinin-chlorophyll-protein from Amphidinium carterae (PCP). It has a wide absorption spectrum (420-550 nm), optimized for sunlight. The dynamics of this protein, used in modern nanotechnology has been not addressed yet. In this work we present results of PCP computer modeling using a well established molecular dynamics methodology. The CHARMM27 force field parameters were prepared for this protein and all chromophore components. The system was embedded in a box of water, with proper counter ions, and a number of 10 ns molecular dynamics simulations were run using the NAMD code. It has been found that peridinine chromophores exhibit substantial orientational flexibility but a pair Per612 and Per613 is more rigid than the remaining two carotenoids. Orientation and dynamics of absorption and emission electric dipole moments have been also analyzed. Apparently, the architecture of PCP is not optimized for efficient Per-Chl a energy transfer by the Förster mechanism. Several practical issues related to molecular dynamics simulation of similar hybrid nanostructures are discussed.

We study the effect of plasmon excitations in silver island film on the optical properties of peridinin-chlorophyll-protein light-harvesting complex using scanning fluorescence microscopy. With this technique we can unambiguously locate areas where the biomolecules are deposited on the metallic nanostructures from the areas where they stick to the glass surface. The enhancement factor of fluorescence intensity obtained for such a hybrid nanostructure is found to be 3. Plasmon excitations in the SIF layer also influence the dynamics of the emission, but in this case the interpretation of the results is more complex.

Eleven proteins belonging to photosystem II (PSII) bind photosynthetic pigments in the form of thylakoid membrane-associated pigment-protein complexes. Five of them (PsbA, PsbB, PsbC, PsbD and PsbS) are assigned to PSII core complex while the remaining six (Lhcb1, Lhcb2, Lhcb3, Lhcb4, Lhcb5 and Lhcb6) constitute, along with their pigments, functional complexes situated more distantly with regard to P680 - the photochemical center of PSII. The main function of the pigment-binding proteins is to harvest solar energy and deliver it, in the form of excitation energy, ultimately to P680 although individual pigment-proteins may be engaged in other photosynthesis-related processes as well. The aim of this review is to present the current state of knowledge regarding the structure, functions and degradation of this family of proteins.

Opis:
Pump-probe spectroscopy after selective excitation of all-trans Cars (n = 9-13) in nonpolar solvent identified a symmetry selection rule of diabatic electronic mixing and diabatic internal conversion, i.e., '1Bu+-to-1Bu- is allowed but 1Bu+-to-3Ag- is forbidden'. Kerr-gate fluorescence spectroscopy showed that this selection rule breaks down, due to the symmetry degradation when the Car molecules are being excited, and, as a result, the 1Bu+-to-3Ag- diabatic electronic mixing and internal conversion become allowed. On the other hand, pump-probe spectroscopy after coherent excitation of the same set of Cars in polar solvent identified three stimulated-emission components (generated by the quantum-beat mechanism), consisting of the long-lived coherent cross term from the 1Bu+ + 1Bu- or 1Bu+ + 3Ag- diabatic pair and incoherent short-lived 1Bu+ and 1Bu- or 3Ag- split incoherent terms. The same type of stimulated-emission components were identified in Cars bound to LH2 complexes, their lifetimes being substantially shortened by the Car-to-BChl singlet-energy transfer. Each diabatic pair and its split components appeared with high intensities in the first component. The low-energy shifts of the 1Bu+(0), 1Bu-(0) and 3Ag-(0) levels and efficient triplet generation were also found.
